Working group·shortcut PP:ANALYTICAL

This page documents a PeptidePedia Wiki working group. It is maintained by the editors who work in this area. Shortcut: PP:ANALYTICAL.

Coordinating chromatography, spectrometry and compendial testing articles.

The analytical working group covers separation science, mass spectrometry, the compendial tests and method validation, and it owns the wiki's treatment of what a purity figure does and does not mean.

Scope

Chromatography, mass spectrometry, water determination, endotoxin and sterility testing, impurities and counterions, reference standards, and analytical method validation. The certificate-of-analysis articles are shared with the quality group.

The standing problem

The group's recurring work is correcting the conflation of area percent purity with peptide content — in articles, in talk-page arguments and in the sources the wiki cites. Any article that quotes a purity figure should make clear what the figure is relative to, and any article that quotes a mass should make clear whether it is gross or net.

Current priorities

  1. Individually source the interference claims in the water-determination articles.
  2. Add a worked limit-of-detection calculation using real signal-to-noise figures.
  3. Document each independent testing service's sampling and reporting practice in its own article, rather than only by comparison with the others.
  4. Standardise chromatographic parameter reporting: column, mobile phase, gradient, detection wavelength, in that order.
